Favoritter
De to største forhåndsfavorittene til den rosa trøyen er Remco Evenepoel og Primoz Roglic. Begge to har hatt en meget god start på sesongen og begge har vis at de kan vinne tre ukers etapperitt tidligere.
I fjorårets Vuelta a Espana var det også disse to som kjempet om seieren inn i den siste uken av rittet. Evenepoel hadde skaffet seg et forsprang, men begynte å se sliten ut mot slutten. Det kunne ha blitt spennende, men endte med at Roglic krasjet og brøt rittet på etappe 16. Nå blir det omkamp mellom de to.
Av andre utfordrere finner vi Tour de France-vinneren fra 2018 Geraint Thomas og lagkamerat og Giro-vinneren fra 2020 Tao Geoghegan Hart. I oppkjøringsrittet Tour of the Alps var det Geoghegan Hart som var den sterkeste av de to.
Portugisiske Joao Almeida, Aleksandr Vlasov og Jack Haig er andre outsidere til en pallplass sammenlagt.
